Progress with Parkinson's!

A naturally-occurring compound has been found to block the molecular process thought to lie behind Parkinson's Disease and could form the basis of a possible treatment, scientists have said.

The preliminary findings suggest the compound, called squalamine, also suppresses the toxic products associated with the process, researchers at Cambridge University have found.
Academics stressed further research is needed, and that findings are based on cell cultures developed in the lab and testing in nematode worms.

But the compound has been used in clinical trials for cancer and eye conditions in America, and a trial in Parkinson's Disease patients is now being planned by one of the researchers involved in the study.
The study was led by academics from the Centre for Misfolding Diseases based at Cambridge University and Georgetown University and the National Institutes of Health in the United States.

It is not yet clear whether squalamine can reach the specific regions of the brain where the main molecular processes determining Parkinson's Disease take place, or what form any resulting drug might take.
Squalamine is a steroid which was discovered in the 1990s in dogfish sharks, although the form now used by scientists is a safer, synthetic analogue.
To date, it has been extensively investigated as a potential anti-infective and anti-cancer therapy.
In the new study, researchers found squalamine inhibits the early formation of toxic aggregates of the protein alpha-synuclein - a process thought to start a chain reaction of molecular events eventually leading to Parkinson's Disease - and can suppress the toxicity of these poisonous particles.
Co-author Michele Vendruscolo, of Cambridge University, said: "This is an encouraging step forward in our efforts to discover potential drugs against Parkinson's Disease."
Researchers said it would be interesting to investigate the efficacy of squalamine as a means to alleviate certain symptoms, including severe constipation and symptoms concerning the peripheral nervous system.
The findings are published in Proceedings Of The National Academy Of Sciences (PNAS).

New DNA mapping project to discover causes of MND launched

Today sees the formal launch in Ireland of Project Mine, an ambitious international effort to find the genetic causes of motor neurone disease. It will involve scientists in 14 countries, including Ireland, who will collect patients’ DNA, sequence it and then do comparative studies that will help provide answers about this terrible disease.

Ireland sees 110 new cases of motor neurone disease every year, says Prof Orla Hardiman, who will lead Project Mine activity in Ireland. Scientists are still learning about the disease, but there is undoubtedly a genetic component, hence the need for an initiative such as this.

“The project is based on looking at the population genetics of motor neurone disease; whole genome association studies, ” says Hardiman, a professor of neurology at Trinity College Dublin and a consultant neurologist at Beaumont Hospital. “It is based on looking at genes, but being able to do this is all about computers, maths and statistics.”

“Ireland is a really good place to do this work; it is good for gene research. It can be easier to do these studies in Ireland because we have larger families and can compare family genes against the wider population.”

The complexity of the genetic background here is also lower than in a country such as the US or mainland Europe.

Only an international effort could tackle such a huge job, says Dr Russell McLaughlin, a postdoctoral research fellow in Trinity’s academic unit of neurology.The genetic resource created by the project will be important not just for motor neurone disease but for research into other conditions. McLaughlin is also involved in a study of irritable bowel syndrome, and the genetic information could be used to study multiple sclerosis, diabetes or Parkinson’s, says Hardiman.

Members of the consortium will share data freely, and so, although we will only pay for a part of the project, we will benefit from having access to all of the resultant data. “We get the data back,” says Vajda. “It is not giving genetic information away, it is bringing it back to us. It represents a new era in gene research. We need to be be involved. We need to be a partner in this to avoid missing out.”

NUI Galway to host major international conference on brain disorders


A major conference on neurodegenerative diseases takes place in Galway this week from 27-28 November.
NUI Galway will host the 24th Annual Meeting of the Network of European Central Nervous System Transplantation & Restoration (NECTAR). The NECTAR meeting is a major international gathering for researchers working on the development of cell and gene therapies for neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease and Huntington’s disease.
In one of the sessions, Professor Roger Barker of the University of Cambridge and one of his patients, Mrs. Sheila Roy, will share their experiences of a new experimental drug called ProSavin® with the audience. ProSavin® is a viral gene therapy manufactured by Oxford BioMedica that is currently undergoing clinical trial for the treatment of Parkinson's disease. It is injected directly into the brain, inducing production of the neurotransmitter dopamine which is deficient in this condition. Trials in laboratory animals and humans have been a success, with ProSavin® demonstrating significant efficacy without the side effects associated with other current treatments for Parkinson's disease.
This conference has been held annually across Europe since its inauguration in Lund, Sweden in 1984, and this is the first time in its history that this prestigious international meeting has come to Ireland.

Network of care improving outcomes for sufferers of Parkinson’s disease


A new approach to care is transforming the lives of patients with Parkinson’s disease and has promise for other long term conditions like COPD and rheumatoid arthritis, according to Dutch researchers writing in the BMJ.

Evidence suggests it empowers patients, improves the quality of care, shifts care away from institutions and into the community, and lowers healthcare costs.

Parkinson’s disease is a common and disabling neurodegenerative disorder. Most patients live with the disease for many years and the burden on patients and carers is considerable.

ParkinsonNet is a model where care is delivered by a network of specially trained professionals who collaborate through a dedicated online platform to which patients also have access. It was developed to tackle concerns of insufficient training for health professionals and poor communication between them and their patients.

Through the platform, patients can find information about treatment options and the trained professionals they need to help them. They are also given the option of having consultations in their own homes through secure video links.

Since its introduction in 2004, ParkinsonNet has now expanded to 66 regional networks and almost 3000 trained experts from 15 different disciplines covering the whole of the Netherlands.
